Recombinant Human TGF-¦Â1/TGF-beta 1, CYT-P01137L

Product Introduction

Transforming Growth Factor ¦Â-1 (TGF-¦Â1) is a secreted protein belonging to the TGF-¦Â family. TGF-¦Â1 is abundantly expressed in bone, articular cartilage, and chondrocytes, and is increased in osteoarthritis (OA). TGF-¦Â1 performs many cellular functions, including the control of cell growth, cell proliferation, cell differentiation, and apoptosis. The precursor is cleaved into a latency-associated peptide (LAP) and a mature TGF-¦Â1 peptide. TGF-¦Â1 may also form heterodimers with other TGF-¦Â family members. It has been found that TGF-¦Â1 is frequently upregulated in tumor cells. Mutations in this gene result in Camurati-Engelmann disease.

Product Features

1. Mammalian Expression System: Produced in a mammalian expression system for proper folding and post-translational processing.

2. High Purity: Greater than 95% as determined by reducing SDS-PAGE.

3. High Biological Activity: ED50 of 4-40 pg/mL as measured by inhibition of IL-4-dependent proliferation of TF-1 human erythroleukemic cells (QC verified).

4. Lot-to-Lot Consistency: Three independent lots tested for activity and plotted on the same graph to demonstrate consistency.

5. Low Endotoxin: <0.01 EU/µg as determined by LAL test.

Storage and Stability

Shipping Conditions: Shipped at ambient temperature. Upon receipt, store immediately at the temperature listed below.

Storage Conditions:

- Lyophilized protein: Store at ¡Ü -20¡ãC, stable for one year after receipt.

- Reconstituted protein solution: Store at 2-8¡ãC for 2-7 days.

- Aliquots of reconstituted samples: Stable at ¡Ü -20¡ãC for 3 months.

Protocol (For Reference Only)

Important: Always centrifuge tubes before opening. Do not mix by vortex or pipetting. It is not recommended to reconstitute to a concentration less than 100 µg/mL.

1. Reconstitution: Dissolve the lyophilized protein in distilled water. Allow to dissolve completely with gentle mixing; do not vortex.

2. Aliquoting: Aliquot the reconstituted solution into single-use volumes to minimize freeze-thaw cycles.

3. Storage: Store aliquots at ¡Ü -20¡ãC for up to 3 months. Reconstituted solution may be stored at 2-8¡ãC for 2-7 days.

4. Activity Assay: Biological activity is measured by inhibition of IL-4-dependent proliferation of TF-1 human erythroleukemic cells, with an ED50 of 4-40 pg/mL.

5. Working Concentration: Optimal working concentrations should be determined empirically for each cell type and experimental system.

Precautions

1. Always centrifuge tubes before opening.

2. Do not mix by vortex or pipetting; gentle inversion or swirling is recommended.

3. It is not recommended to reconstitute to a concentration less than 100 µg/mL to avoid protein loss.

4. Aliquot reconstituted solution to minimize freeze-thaw cycles.

5. For research use only. Not for use in diagnostic or therapeutic procedures.

FAQ (Simplified)

Q1: How should I reconstitute this protein?

A1: Dissolve the lyophilized protein in distilled water to a concentration of at least 100 µg/mL. Do not vortex or pipette vigorously.

Q2: What is the biological activity of this protein?

A2: The ED50 is 4-40 pg/mL as measured by inhibition of IL-4-dependent proliferation of TF-1 human erythroleukemic cells.

Q3: How should I store the reconstituted protein?

A3: Reconstituted solution can be stored at 2-8¡ãC for 2-7 days. For longer storage, aliquot and store at ¡Ü -20¡ãC for up to 3 months. Avoid repeated freeze-thaw cycles.

Q4: What is the endotoxin level?

A4: <0.01 EU/µg as determined by LAL test.
