Product Introduction

TAE (Tris-Acetate-EDTA) buffer is a commonly used electrophoresis buffer for DNA gel electrophoresis. The 1¡Á working solution contains 40 mM Tris-acetate and 1 mM EDTA, pH 8.0. TAE is suitable for the separation of DNA fragments in agarose gels, particularly for fragments larger than 1 kb and for applications requiring DNA recovery for downstream enzymatic reactions. Compared to TBE buffer, TAE has lower buffering capacity but provides better DNA recovery and is preferred for preparative gels and applications where DNA will be purified for cloning or sequencing. This product is supplied as a 50¡Á concentrated stock solution and should be diluted 50-fold with distilled water before use.

1. Preparation of 1¡Á Working Solution: Dilute the 50¡Á TAE buffer 1:50 with distilled water (e.g., 20 mL of 50¡Á TAE plus 980 mL of distilled water to prepare 1 L of 1¡Á TAE). Mix thoroughly.

2. Agarose Gel Preparation: Dissolve agarose in 1¡Á TAE buffer by heating. Cool and cast the gel.

3. Electrophoresis: Submerge the gel in 1¡Á TAE buffer. Load DNA samples and run at appropriate voltage.

4. Buffer Reuse: 1¡Á TAE buffer may be reused for multiple gel runs, but prolonged reuse may reduce resolution. Replace buffer when bands become diffuse or resolution decreases.

Precautions

1. If precipitation occurs during storage, warm to room temperature and mix until fully dissolved.

2. Do not use the 50¡Á concentrate directly for electrophoresis; always dilute to 1¡Á.
