Introduction

The N-2 Supplement (100X) is a defined, concentrated serum-free supplement designed for the growth and maintenance of neuronal cells and neural stem cells in culture. It provides a mixture of essential components, including hormones, proteins, and other factors, that support the specialized needs of these cell types, eliminating the requirement for serum and thus providing a more controlled environment.

Intended Usage

This 100X concentrate is intended to be added at a 1:100 dilution to an appropriate basal medium for the *in vitro* culture of primary neurons, neuroblastoma cell lines, PC12 cells, and neural stem/progenitor cells.

Kit Components

-N-2 Supplement (100X): 5 mL / bottle;10 mL / bottle. 

-The supplement is supplied as a sterile liquid.

Storage

-Upon receipt, store the N-2 Supplement (100X) at -20¡æ.

-Protect from light.

-Aliquoting is recommended to avoid repeated freeze-thaw cycles.

-Once thawed, the supplement can be stored at 2¡æ to 8¡æ for up to 2 weeks. Do not refreeze.

Composition

The N-2 Supplement is a defined formulation containing:

-Human Transferrin (Holotransferrin)

-Insulin (Recombinant Human)

-Progesterone

-Putrescine

-Selenite (Sodium Selenite)

Preparation of Complete Medium

1.  Thaw the required amount of N-2 Supplement (100X) completely. Mix gently.

2.  Aseptically add the N-2 Supplement to your pre-warmed basal medium (e.g., DMEM/F12, Neurobasal™ Medium).

3.  The standard recommended dilution is 1:100.

    -Example: To prepare 500 mL of complete medium, add 5 mL of N-2 Supplement (100X) to 495 mL of basal medium.

4.  Mix the complete medium thoroughly by gentle swirling.

5.  It is recommended to use the complete medium immediately. If storage is necessary, keep it at 2¡æ to 8¡æ, protected from light, and use within one week.

Precautions & Warnings

-For Research Use Only. Not for use in diagnostic or therapeutic procedures.

-This product is not sterile-filtered. It is manufactured under aseptic conditions, but users must employ aseptic technique.

-All biological materials should be treated as potential health hazards. Wear appropriate personal protective equipment (PPE) such as lab coats and gloves.

-Do not use if the solution appears cloudy or contains particulate matter.

Technical Notes

-The optimal concentration of N-2 Supplement may vary depending on the cell type and basal medium used. We recommend testing a range from 0.5X to 2X the standard concentration for your specific application.

-For long-term cultures (>4 days), medium change with fresh N-2 containing complete medium is recommended every 2-3 days.

Quality Control

Each lot of N-2 Supplement (100X) is functionally tested for its ability to support the growth of specified neuronal cell lines.
